My current research focus is the integration of multi-dimensional genomics datasets associated with human disease, with particular emphasis on the genetic aetiology of complex neurological disorders. I am developing methods for reference-based and reference–free genome analyses to identify known and novel genomic variation of predicted functional relevance. Our analyses integrate short-read and long-read technologies to inform assemblies and reveal cryptic alterations that are generally missed by short read sequencing methods. The primary focus of my projects is to investigate the genetic biomarkers and pathogenic mechanisms of neurological disorders, with an initial emphasis on dystonia, Parkinson’s disease and X-linked Dystonia Parkinsonism (XDP), and planned expansion into other neuro-degenerative and neuro-developmental disorders. The pathogenesis of neurological disorders often involves multiple genes, complex interactions thus, the functional genomics aspect of my project employs genome-wide and targeted transcriptomics analysis of post-mortem brains from different neurological disorders to investigate common pathological mechanisms which concur among these disorders.
